For the more significant part of a decade, I’ve been captivated and consumed with the tremendous complexity of the human immune system.

It started in university when I was studying information design and needed a semester project. The immune system seemed like a suitable fit. So I gathered a large stack of immunology books and dove in, but things remained confusing no matter how much I read.

Since I gained knowledge, it became increasingly challenging to simplify the immune system, as each layer revealed additional mechanisms, exceptions, and complexity. As a result, a project scheduled to take only until spring ended up taking over the summer, then the fall and winter. The interconnections between the immune system’s components were too graceful, and the dance they performed too exquisite to stop learning about them. This progression drastically altered my perceptions and feelings about my body.

As soon as I got the flu, I couldn’t just complain. I had to look at my body, touch my swollen lymph nodes, and visualize what my immune cells were doing at the time, which section of the network was active, and how T Cells killed millions of intruders to protect me. When I cut myself in the wilderness while being reckless, I felt grateful for my Macrophages. These giant immune cells chase fearful bacteria and tear them apart to protect the open wound from infection.

It occurred to me while in the ICU that mast cells and IgE Antibodies had nearly killed me in their mistaken attempt to protect me from scary foods, so I started thinking about them!

When I was diagnosed with cancer at thirty-two and required many operations and subsequent chemotherapy, my preoccupation with immunology intensified. One of my immune system’s functions is to eliminate cancer.

It had failed in this instance. But I couldn’t be furious or outraged because I had learned how difficult this task was for my immune cells and how difficult it was for cancer to keep them in check. And as the chemo dissolved cancer, my thoughts returned to my immune cells infiltrating and devouring the dying tumors one by one.

Disease and illness are frightening and disturbing, and I’ve encountered plenty of both throughout my life. However, knowing how my cells, immune system, and other parts of myself protect the thing that is me, how it fights and dies, and how it heals and repairs the body I live in has always given me a lot of comforts. Learning about the immune system has made my life better and more exciting, and because of this, there is less fear of getting sick.

Understanding the immune system always helps to put things into perspective. As a result of this beneficial effect, and just for the enjoyment of learning and reading about the immune system, it became a continuous passion, especially as I later became a science communicator, and explaining complicated concepts became my life’s goal. About eight years ago, I founded Kurzgesagt—In a Nutshell, a YouTube channel dedicated to simplifying and beautifying information while being as loyal to science as possible.

By early 2021, the Kurzgesagt crew had grown to over forty members, with the channel having accumulated over fourteen million subscribers and reaching about thirty million monthly viewers. Therefore, if such a vast platform exists, why endure the agonizing process of publishing this book? While some of our most popular films have focused on the immune system, it has always bothered me that I was unable to cover this fascinating subject in the detail it deserves. A ten-minute video is not the appropriate medium for such an undertaking.

Even if you overcome these obstacles and continue your quest to understand the immune system, you will run upon another stumbling block: the humans who described it. Scientists worked hard and were always curious to make the world we live in today even better. We owe them a debt of gratitude. Unfortunately, many scientists are terrible at selecting appropriate names and developing accessible language for what they uncover.

Immunology is one of the most egregious offenders of any scientific discipline in this regard. Primary Histocompatibility Complex class I and II, gamma delta T cells, interferon-alpha, beta, gamma, kappa, and the complement system, which includes parts called the C4b2a3b complex, make a field that was already complex even more so.

Thus, the purpose of this work is to do a delicate dance around all of these issues. It will communicate in plain English and utilize complex terms only when necessary.

Processes and interactions will be simplified as possible while remaining true to the science. The complexity of each chapter will gradually increase so that after you’ve been inundated with knowledge, there will be more laid-back sections to unwind. And at regular intervals, we shall synthesize what we have learned.

I want this book to enable anyone to understand their immune system and have some fun while doing so. I should point out that I am not an immunologist but a science communicator and a fan of the immune system. This book will not satisfy every immunologist—as was evident from the start, there is much debate among the experts who hold these theories, which is precisely how science should function!

For instance, some immunologists regard particular cells as irrelevant fossils, while others believe they are necessary for your defenses. As a result, this book is mainly based on interactions with scientists, current immunology literature used in classrooms, and peer-reviewed articles. Nonetheless, portions of this book will require updating in the future.

Dettmer uses simple metaphors to show how hard-to-understand words are offered so that even a person who doesn’t know the language can understand what is going on. In this example, MHC Class II receptors, antigens, and MHC I molecules are all hotdogs. MHC Class I molecules are like doorways into the cell’s machinery. These simple diagrams are beneficial, and the illustrations use the Kurgzesagt channel’s great ideas. In addition, I found the footnotes to be a great way to explore interesting side stories while keeping the main plot (if you can call it that) on track.


In the middle of Dettmer’s talk about immune pathways, some of his ideas start to lose their focus. He has an intensely reductionist lens that only looks at paths. This makes it hard for him to see the whole picture. When it comes to immunology, this is how every textbook and class I’ve come across deals with the subject. This is not just Dettmer’s fault. But it leads to a very strange cyclical logic that is hard to understand.
